Various genetic-based approaches including mutant population screens, microarray analysis, cloning and transgenesis have expanded our knowledge of gene function during meiosis in plants. However, this genetic tool is not without a limitation attached. One alternative approach to studying meiosis plants, especially in polypoid such as Triticum Aestivum L. (wheat bread), is proteomic.
However, protein-based approaches using proteomics are rarely explained, with only two efforts to study the initial plant meiosis reported. Here, we report an inquiry of Meiosis wheat early bread using proteomics. Five protein spots that differed differently were identified using 2D gel electrophoresis (2DGE) in protein extract from four stages of Pound Meiosis and three genotypes (Chinese spring wild type, PH1B and PH2A wheat mutants). Tandem Mass Spectrometry (MS / MS / MS) Identification of peptides from these protein spots causes insulation and characterization of full length clones of wheat speckle type Puze proteins, proteins such as SF21 and HSP70, and the order of partial coding.
Hexose transporter. Significantly, the pale function of the protein POZ type Speckle and HSP70 is confirmed using the DNA binder test in vitro. Through the use of a 2DGE proteomic approach, we show that proteomic is a feasible alternative for genetic-based approaches when studying meiosis in wheat. More significant, we report a potential role for speckle type PUZ proteins and HSP70 in chromosome pairs during the early stages of meiosis on wheat bread.
Assessment of genotoxic effects of pendiumphalin in Chinese hamsters over cells with a single cell electrophoresis test (comet)
PendiMethalin (N-ethylpropopyl) -3.4-dimethyl-2,6-dinitrobenzenine) is a dinitroaniline herbicide compound that selectively controls weeds. This is cell division and growth inhibitors. It goes down the plant in a short time after Seedling. This is a soil and water pollutan because of the widespread use of formulations in Turkey and throughout the world. PendiMethalin is produced and imported by Turkey. PendiMethalin is a slightly toxic compound that is classified in class 3 toxicity by the United States Environmental Protection Agency (USPA). Although classified as Group C (human maybe carcinogens) compound by USEPA, there are a number of studies of their genotoxic effects. The purpose of this study was to evaluate vitro genotoxic effects from various methalized concentrations in the top Chinese hamsters (CHO) cells with a single cell gel electrophoresis test (comet).
Materials and Methods: Cells incubated with a concentration of pendinalin 1, 10, 100, 100, 1000 and 10000 μm for 30 minutes at 37 ° C and DNA damage compared to Cho cells that are not treated with pendiMethaline. 50 μm hydrogen peroxide is used as a positive control.
Results: There are no significant cytotoxic effects observed in the concentration range studied. DNA damage in Cho cells increased significantly in the concentration of pendiDalinalin 1, 100, 1000 and 10000 μm, a significant decrease was observed in the concentration of 10 μm pendiumphalization.
The application of capillary zone electrophoresis (CZE) and capillary gel electrophoresis (CGE) for the separation of human insulin, lispro insulin and their degradation products.
The method of two capillar electrophoresis (CE) has been developed for the separation of cars and mass variants of human insulin and recombinous analogues of Lispro. Because the capillar zone (CZE) and capillar gel electrophoresis (CGE) are based on different separation principles, they can be used to detect different insulin impurities and analogues. The application of CZE allows the separation of compounds with different M / Z ratios, therefore CZE is a suitable method for the separation of Deamidation Insulin products. After optimization, this method is validated according to the requirements of ICH. The CGE method is used for separation of higher molecular weight transformation products. Experimental data shows that CZE and CGE are simple, fast and strong methods that can be used as routine analysis for the quality control of insulin formulation

Production of high-quality two-dimensional gel electrophoresis profiles for MEDAKA Marine samples using trizol-based protein extraction approaches.
Medaka Marines is one of the most popular species of fish species for ecotoxicology and environmental research and proteomic studies is a tool that is useful for understanding Medaka molecular responses on exposure with environmental stressors. The preparation of high-quality protein samples is the key to producing high-quality two-dimensional electrophoresis gel results (2-DE) for proteomic analysis. In recent years, trizol-based protein extraction has gained popularity because of its promising performance in producing high-quality 2-de and convenience methods.
Three Trizol-Based Approaches (Trizol Methods, Trizol Alikuot Methods and Trizol Methods with Commercial Cleaning Kits) are used to extract proteins from Medaka Marine samples and 2-de profiles produced. 2-de profile quality and effectiveness of extraction methods evaluated. In comparison, two common protein extraction methods (methods of buffer lysis and extraction of trichodic acid / acetone precipitation) are also applied in parallel with a trizol-based approach. Each trizol-based approach produces high-quality 2-de high-quality Marine Profile Medaka compared to the two methods of Buffer Lisis and TCA / Aceton rainfall extraction.
